Anti-CD20 Antibody Comparison: Rituximab Biosimilars
About CD20 and Rituximab biosimilars
CD20 (also known as MS4A1) is a B-lymphocyte surface protein expressed on cells from late pre-B-cell through to memory B-cell stages of differentiation. It does not appear on plasma cells or early stem cells, which is why it has become a high-value target for monoclonal antibody therapies in B-cell lymphomas, chronic lymphocytic leukaemia and autoimmune disorders.
Rituximab is the first-in-class anti-CD20 monoclonal antibody, a chimeric mouse/human IgG1 originally developed by IDEC Pharmaceuticals and marketed as Rituxan® (Genentech/Roche). It binds a large extracellular loop of CD20 and triggers B-cell depletion through complement-dependent cytotoxicity (CDC), antibody-dependent cellular cytotoxicity (ADCC), and direct apoptosis.
Research-grade Rituximab biosimilars are recombinant antibodies that incorporate the same variable region as the therapeutic Rituximab, formatted for laboratory use. They are not clinical-grade and not approved for human treatment, but they are valuable for biosimilar comparability studies, CDC and ADCC functional assays, CAR-T construct validation, and B-cell research generally.
This page compares the four Anti-CD20 Rituximab biosimilars with publicly listed catalog numbers from major suppliers. They share the Rituximab variable region but differ in host species, format and which applications each vendor has validated. The matrix below shows facts; the buyer applies their own criteria.

Validated Applications for Each Anti-CD20 Antibody
Which products are documented as validated for which application. Sourced from each vendor's product page. A check means the vendor lists that application; a dash means it is not listed (which is not the same as not working; the vendor simply hasn't validated for it).

Choosing the Right Anti-CD20 Antibody for Your Workflow
These rules teach you which column in the table to weight. They do not pick a product for you.
- In-vivo or murine model workCompare the Host species and Format columns. Mouse-host and chimeric biosimilars behave differently in murine models than human-host recombinants.
- Clinical-workflow IHC on FFPE tissueCompare the Applications column for IHC-P validation and the Host species column. Rabbit-host antibodies are often preferred for IHC-FFPE staining workflows.
- Functional assays (CDC, ADCC, neutralisation)Compare the Format and Applications columns. Low-endotoxin or LEAF-formatted products avoid confounding effects in functional readouts.
- Published validation depthCompare the Citations column. Higher citation counts mean more peer-reviewed publications using that exact product, which reduces methodological risk.
- Multi-application validation in one antibodyCompare the Applications column for the longest validated list. A single antibody validated across more applications reduces SKU count and improves cross-experiment comparability.
